Image Annotation: ES Targeted Insertion; K-ras latent allele is a targeted insertion of a vector carrying K-ras exon 1 with an activated codon 12 mutation (gly-asp). This insertion results in a duplication of exon 1 sequences separated by plasmid sequences and the neomycin resistance gene. This mutation was introduced into 129/sv embryonic stem(ES) cells. In the germline configuration, the mutatnt allele is expected to be non-functional and is thus carried in the heterozygous state. Due to the close proximity of these duplicated sequences, an oncogenically activated allele of K-ras spontaneously occurs via intrachromosmal recombination or sister chromatid exchange events (rate unknown).
